چکیده

Linked Soft Gelatin Capsules S. Tindal, K. Flood, J-E. Early, L. Covington, Z. Karjoo, E. Bailey Catalent Pharma Solutions Purpose USP <711> Dissolution is under revision to update the Tier 2 testing parameters for gelatin capsules where cross-linking of the gelatin has been observed and is impacting the dissolution results of the gelatin capsule. The revision introduces two new enzymes for use in Tier 2 testing over the pH ranges where the current enzymes are not effective in cleaving the cross-linked gelatin to enable rupture and disintegration of the cross-linked capsule. The new enzymes, papain and bromelain, are included for use in dissolution media with pH values ranging from 4.0 – 6.8. The revision also changes the pH range where pepsin is used from 1 – 6.8 to 1 – 4, increases the amount of pancreatin from 1750 UPS units / L to 2000 USP units / L and provides guidance for the use of an enzyme pre-soak for media where enzyme activity is inhibited (i.e., media containing some surfactants).

Dissolution of Gelatin Capsules: Evidence and Confirmation of Cross-Linking

Cross-linking is a common problem in the dissolution of gelatin capsules. Cross-linking is characterized by a bridge across the peptide backbone of the gelatin molecule which creates water insoluble membranes or pellicles during dissolution testing. The chemical covalent bonding between gelatin chains is typically triggered by catalytic amounts of aldehyde and/or from exposure to high temperatu...

A Review of the Problem, Test Methods, and Solutions

www.pharmtech.com anufacturers of pharmaceutical products have the responsibility — not only from marketing and ethical standpoints but also from a legal (regulatory) perspective — to ensure that their products meet dissolution specifications during storage conditions described on the label. This is necessary because dissolution per se is rate determining in terms of the absorption and bioavail...

Inverse Miniemulsion Method for Synthesis of Gelatin Nanoparticles in Presence of CDI/NHS as a Non-toxic Cross-linking System

In this research, gelatin nanoparticles were synthesized via inverse miniemulsion method by employing a mixture of a water soluble carbodiimide (CDI) and N-hydroxysuccinimide (NHS) as a non-toxic cross-linking system. The gelatin nanoparticles were characterized for their size and size distribution, morphology and stability and were compared with those of nanoparticles cross-linked by glutarald...

Effect of Some Synthetic Parameters on Size and Polydispersity Index of Gelatin Nanoparticles Cross-Linked by CDI/NHS System

In our previous work, the effect of use of a water soluble CDI/NHS system as nontoxic cross-linking agent on fabrication of gelatin nanoparticles was investigated. In this research, the effect of variation in some synthetic parameters of gelatin nanoparticles cross-linked by CDI/NHS system such as type of gelatin and formulation of cross- linking agent on their size and distribution was examine...

Crosslinking Of Gelatin Capsules

~emistry of the crosslinking of gelatin is a complex phenomenon. the nature of which is dependent on several factors. The present manuscript represents an attempt to answer some specific basic science issues. Two "stress" conditions are commonly known in connection with gelatin capsules. The first condition is induced by exposure to formaldehyde. The second stress condition is created by exposi...
